Context
Cell therapies are advancing, but current approaches depend on the microenvironment. Leukotroie aims to dynamically control the reactivation of immune cell functions to enable new antitumor strategies and/or promote tissue regeneration.
Technology
Optogenetic technology acting within immune cells. Precise, specific, and reversible control of intracellular signaling pathways, particularly the Src family, which is central to macrophage activation and modulation of their effector functions.
Advantages
Technological breakthrough: intracellular rather than extracellular action. Precise spatiotemporal control of cellular functions. Stereotyped and reproducible responses. Targeted induction of antitumor activities, with potential conversion of “cold” tumors into “hot” tumors, or pro-proliferative effects, using optogenetic versions of Src kinase.
State of progress
Modular optogenetic construct portfolio: OPTO-LYN (antitumor activity) and OPTO-SRC (proliferation/tissue repair). Activities validated in cell spheroids. Platform versatility demonstrated across multiple indications. In vivo results expected.
Applications
Applications in immuno-oncology for the treatment of solid tumors (melanoma, pancreatic, lung cancers, etc.) through activation of antitumor responses and modulation of the tumor microenvironment. Also applicable to tissue regeneration and repair.
