Context
Tumoroid models derived from patients currently represent a promising alternative. They enable better reproduction of tumor heterogeneity and cell-cell interactions in a three-dimensional structure. However, current approaches have several limitations.
Technology
The TOTEM project aims to transform the treatment of aggressive cancers using a unique ex vivo tumoroid technology generated immediately from a fresh tumor fragment. This device makes it possible to recreate a tumor avatar that retains its 3D architecture in just a few minutes.
Advantages
Speed: usable results in ≤7 days
Accuracy: preservation of 3D architecture, microenvironment, and rare cell populations
Simplicity: ready-to-use kit
Scalability: consumable, standardized, and reproducible format
State of progress
The development of TOTEM benefited from a series of validations carried out in a variety of environments. Viable tumoroids were established on 88 brain tumors. The design of a functional prototype incorporating a nanoporous silicon support was consolidated.
Applications
. Uses in R&D and creation of tumoroid biobanks
. Pharmaceutical development
. Clinical application in personalized medicine
